Hematoxylin (Hematoxylin) and Eosin (Eosin) combined staining referred to as HE staining, is the most commonly used method of staining in pathology and histology, Hematoxylin is an alkaline natural dye, which can color the nucleus of cells; the main component of chromatin in the nucleus of the cell is DNA, and in the double-helix structure of the DNA, the phosphoryl groups on the two nucleotide strands are outwardly oriented, so that the outer side of the double helix of DNA is negatively charged and acidic. In the double helix structure of DNA, the phosphate groups on the two nucleotide strands are outwardly oriented, which makes the outer side of the DNA double helix negatively charged and acidic, and it can easily be stained by combining with the positively charged hematoxylin alkaline dyes by ionic or hydrogen bonds.

Modified Lillie-Mayer hematoxylin staining solution is non-toxic, no oxidized membrane, the chromatin of cell nucleus is stained deeply and finely, and is often used as a substitute for Harris hematoxylin staining solution in clinic; the staining of the cell nucleus is very clear, and it does not adhere to the staining of cytoplasmic and fibrous components, and it is a progressive staining, so the staining can be done without the hydrochloric acid ethanol differentiation, and the staining time is about 5-8min, and the dyeing time is about 3-5min, and if it is oxidized sufficiently, it can be stained for 3~5min. The reagent is often used in glycogen and other special staining, enzyme histochemistry and immunohistochemistry and other staining of the cell nucleus for control staining, especially for special staining can not be treated by acid staining of the nucleus of the cell, in special staining is often used in conjunction with the azurite blue B liquid staining, so that the nucleus of the cell staining is not followed by the fading of the acidic dyes.
