The staining solution mainly consisted of Alcian stain (PH2.5) and nuclear solid red re-staining solution. The staining result is: acid mucin is blue, proteoglycan and hyaluronic acid are blue, and cell nucleus is red.

Alisin blue staining solution is suitable for staining carbohydrates, such as acid mucin, proteoglycan, hyaluronic acid, etc., but it can't distinguish between proteoglycan and hyaluronic acid. pH2.5 is more suitable for staining saliva-acidic mucus substances (or hydroxyl-containing mucus substances), the new cryptococcal staining, and pH1.0 is more suitable for staining sulfur-containing mucus substances.
