RELA-C11Orf95 fusion-positive ventricular meningiomas are prevalent in children and young adults and are located supratentorially (accounting for more than 70% of supratentorial ventricular meningiomas), characterized by the presence of chromosome 11q13.1 fragmentation and rearrangement to form a RELA-C11Orf95 fusion and the high expression of the cellular adhesion molecule L1CAM, and histologically equivalent to a grade II ventricular meningioma or a grade III interstitial metaplastic ventricular meningioma. The prognosis is worse than other supratentorial ventricular meningiomas, with a 10-year progression-free survival rate of less than 20%;


Because RELA and its partner gene C11ORF95 are only 1M away from each other in the genome, if the region covered by the fragmentation rearrangement of 11q13.1 is small, the red and green signals of the RELA breakage probe may not be separated, and it is difficult to recognize the negative/positive cells under the microscope; if the region covered by the fragmentation rearrangement is large, the red and green signals of the RELA breakage probe may be obviously differentiated, and it is relatively easy to read under the microscope; the complexity of chromatin fragmentation rearrangement is the same as the signal of the positive cells. The complexity of chromatin fragmentation and rearrangement is positively correlated with the complexity of the positive signals.The interpretation of RELA breaks detected by FISH is difficult, and it is recommended to combine with PCR when encountering uncertain results.
